Transthyretin (TTR) is a homotetrameric protein causing fatal amyloidoses. Tetramer dissociation causes aggregation and triggers disease onset. Tolcapone is an FDA approved drug that binds specifically to TTR in human blood, stabilizes the tetramer and inhibits TTR cytotoxicity.
