The HIV gp41 ectodomain has a trimeric coiled-coil structure in which each monomer forms a hairpin of two alpha helices connected by a loop. The ectodomain is highly insoluble under physiological conditions. Computational analysis identified sequence regions prone to aggregation (A). Seven residues in the loop where mutated to produce a fully soluble ectodomain (B). A series of reversed mutants where produced and biophysically characterized. Solubility studies of the mutants (C) allowed identifying the role of mutated amino acids on the solubility of the ectodomain. A model for self-association of the ectodomain was proposed (D).
