In human mitochondria, the transcription factor TFAM activates the promoters at the mitochondrial DNA control region. In addition, TFAM compacts this DNA. We show that whilst the binding dynamics of TFAM to the DNA is strongly tuned by DNA physical features, it induces a U-turn in all cases. A concentration effect causes aggregation of TFAM/DNA complexes via specific TFAM surfaces, suggesting a mtDNA compaction mechanism in which U-turns are the compacting unit.
